Description

Echinacoside, a natural polyphenolic compound, has various biological activities, such as antioxidative, anti-inflammatory, neuroprotective, hepatoprotective, nitric oxide radical-scavenging and vasodilative actions. Echinacoside, one of the phenylethanoids isolated from the stems of Cistanche salsa, elicits neuroprotection by activating Trk receptors and their downstream signal pathways. Echinacoside also has a remarkable antiosteoporotic activity.

In vitro

Echinacoside behaves in vitro as an anti-oxidant and a potent free radical scavenger. It has been shown to rescue SH-SY5Y neuronal cells from TNF-alpha-induced apoptosis. Echinacoside significantly attenuates glutamate-induced neurotoxicity and exhibits neuroprotective activities in vitro. Echinacoside has significant neuroprotective effects on TNFα-induced apoptosis in SHSY5Y neuronal cells by maintaining mitochondrial function, decreasing the generation of reactive oxygen species, increasing the expression of the antiapoptotic protein Bcl-2 and inhibiting caspase-3 activity through an antioxidation mechanism.

In vivo

Echinacoside has neuroprotective effects in the mouse MPTP model of Parkinson's disease. Treatment with echinacoside improves motor behavior and attenuated the depletion of striatal dopamine, DOPAC and HVA in MPTP-lesioned mice.
